Annotation: Within the extended Technical contract we will continue to supply of live 10,000 tsetse fly puparia as required to participants of the CRP „Enhancing tsetse fly refractoriness to trypanosome infection“a. 200 puparia shipped from G. m. morsitans, G. pallidipes; G.palpalis gambiensis and G. f. fuscipes colonies monthly to S. Aksoy, Yale. b. Four allotments of 200 puparia from G. m. morsitans shipped to A. Malacrida, Pavia. c. Puparia (1000 for each species) from G. pallidipes and G. fuscipes shipped to N. Maniana, ICIPE. At the same time we set up the collaborative experiments with Aksoy´s lab. concerning the maternal transmission of endosymbiotic bacteria to progeny during the larval intrauterine development. The study will be focused on the bacteria and progeny molecular cross-talk, bacterial transcriptome changes in the adult female and in larvae throughout development. Bacterial gene expession during development will be related to the expression of host molecules that may modulate transmission of trypanosomes in the emerging progeny, particularly those that with anti-trypanosomal activity.

Annotation: The main idea of the project is to build up a backup colony of tsetse flies Glossina pallidipes, which will serve to provide the mass rearing operations of tsetse flies in STEP in Kaliti, Addis Ababa, Ethiopia. These operations will produce large quantities of sterile male tsetse flies, which will be gradually released in the areas of tsetse occurrence, to eradicate them. Method of sterile male application (Sterile Insect Technique-SIT) has been successfully applied to a number of insect pests and is safer, cheaper and more effective than the use of harmful insecticides. SIT is particularly unique in that, the only environmentaly safe method, and when is used consistently, eradicates harmful insects at the exactly limited area.

Annotation: The distribution and genetic population structure of most species were greatly affected by the climatic cycles during Pleistocene glaciations. Populations surviving ice ages in southern or central Europe served as sources for reinvasion during the interglacial times. Few studies investigated the colonization pattern in freshwater systems in Europe. Nevertheless, there is a persisting disagreement according to the investigated fish species. The project suggests to compare colonisation strategies of two species occurring in the same river system but inhabiting different parts of the river stretch. Results will reveal the genetic diversity and population structure of bothspecies and should help to understand the evolutionary process that led to the present distribution of fish gene pools in one of the main river systems of Europe. Obtained information could be useful in elucidating between recent and more ancient evolutionary events that have promoted species diversification.

Annotation: Song of passerines have several functions, like territory defense and mate attraction. By using of reproducedsong of higher and lower quality in the Reed Warbler we will simulate occupied territories by males of differentquality. We will determine the male strategy of territory choice affected by simulating quality of neighbor song, thefemale mate choice in relation to quality of neighbor song and evaluate the maternal investment of females intoeggs in terms of clutch size, egg yolk testosterone and egg white lysozyme, in dependence on quality of neighborsong. Long-distance migrant songbirds are among the most endangered species. Their migration strategies arealmost unknown and their relation to reproductive strategy, population trend and endangering factors is being notexplained sufficiently. Using geolocators we will study the migration strategies and routes of selected individualsof the Lesser Grey Shrike and analyze their breeding success as well local and global factors of endangerement.

The role of ectoparasite-ectoparasite interactions in the transmission of vector-borne parasites

Annotation: One of the biggest challenges faced by ecologists is to understand how and to what extent interspecific interactions within and between different guilds influence community structure, abundance of specific species or biodiversity of organisms of interest such as pathogens. In spite of a general acceptance of the existence of such a complex system, most ecological interactions are studied in relative isolation. The goal of this project is to see how the interactions between a certain host and a certain vector-borne parasite are affected by the complexity of ectoparasite-ectoparasite interactions. We specifically aim to establish if the interactions among multiple blood-feeding dipteran ectoparasites can influence the susceptibility of an avian host to some of the ectoparasites and/or pathogens that they vector. Namely, we plan to reveal if host susceptibility to haemosporidians can be mediated by 1) ectoparasite-ectoparasite interference or 2) modulation of host immunity by background ectoparasite pressure.

Annotation: The human impact on ecosystems and communities of different organisms is highly actual topic. Mankind is changing its environment continuously and is adapting it to its needs without any regard to other biota. By indirectimpact we often unknowingly decrease diversity of all communities (as plants as animal). This way also communities of small soil arthropods are affected that play a serious role in nutrient cycling due to cutting of fallenleafs. This project will be focused on oribatid mite communities in urban habitats and other biotops affected byhuman impact. Oribatids are soil mites and for their communities it typical high diversity in natural ecosystems.This diversity decreases withincreasing human impact and it also changes species composition of thecommunity. Using structure of orbatid mites community we want to define differences between different ways how to care for “green parts” of the city.

Annotation: Saproxylic mites are those animals that are dependent on dead or decaying wood (or dependent on other organisms that are themselves dependent on such life substrates). They are showing various phoretic associations with different bark and woodboring insects, as a result of specific evolutional adaptations and complex life strategies developed in the mites adapted to the life in spatially and temporally ephemeral habitats, including specific arboricolous microhabitats such as bark beetle galleries of trees, subcorticolous detritus, tinder, decaying wood, wood-destroying fungi, a.o. The project will contribute to original knowledge on species diversity, ecological requirements, bionomy, chorology and altitudinal (vertical) distribution of found saproxylic mites occurring in Europe, with original identification keys, descriptions of all recorded taxa, revisions of unclear and not adequately described species, and ecological and zoogeographical evaluations.

Annotation: Emergence and spread of new infectious diseases is becoming a never more serious medical, societal and economical problem. Increasingly more diseases expand their ranges or re-emerge in areas from where theyhad been wiped out long ago. The emergence of new diseases is most frequently attributed to global environmental changes, including climate change, land use changes and changes in human concentration. A key role in the spread of new diseases is attributed to changed patterns of animal migration and therefore also oftheir parasites – potential vectors of diseases. Despite great effort, knowledge into the causes of the successful transmission and spread of vector-borne parasites is still only poorly understood. Consequently, theoreticalprediction models of the spread of diseases and efforts to halt this spread are not effective. The aim of this project is to increase the understanding of the causes of host specifity and the regulation of circulation of vectorborne parasites. We will use two globally important study systems that involve tick-borne Borrelia spirochetes and insect-borne Plasmodium and Haemaproteus haemosporidians. The objective is to identify taxonomic, genetic and phylogenetic structure of local host associations and to assess if host specificity is a result of hostspecialization by parasites or i tis a result of host and vector ecology.

Annotation: Emerging tick-borne encephalitis from May this year in eastern Slovakia, has been the largest in number ofcases for the last five years. As usual, it was supposed that products from raw sheep\’s milk had been thecausative agens. Such enormous epidemic is striking due to the regular routine screening of sheep milk. Wehave known many foodborne infections from history, but most of cases was related to goat\’s milk. The aim of theproposed project is a comparative analysis involving sheep respectively goats foodborne infections. With regardto exact criteria it will be selected a representative number of farms from all Slovakia. We will investigate the milkof sheep and goats, as well as ticks collected from direct observation of the animals. It will be done for eachfarm. Sample will be examined by highly sensitive molecular biological methods for the virus presence. Fromanimal blood we evaluated the serological status of all investigated animals. Evaluation of the results we can seehow engaged sheep and goats in the case of food-borne infections. In the case of virus positive samples bysequencing analysis we obtain very valuable information about the genetic diversity of the TBEV in our country.An equally important outcome of the project will be specific methodological recommendations for farmers interms of preventing foodborne disease.

Annotation: Insects are widespread and evolutionary extremely successful group of animals that show unique adaptations during development and reproduction. These adaptations are mediated by special endocrine glands including corpora allata (CA) that produce juvenile hormones (JH) and various modifications of reproductive organs. These important organs are under control of regulatory compounds that include neuropeptides produced by neurons and endocrine cells. However, neuroendocrine processes controlling activity of the CA in different developmental stages are not fully understood. Our preliminary and published data indicate that JH production from the CA is controlled by several peptidergic cells, whose specific functions in this process remain to be determined. Likewise, the role of neuropeptides during mating, fertilization and egg laying is not fully understood. Therefore, in this project we will identify neuropeptides, receptors and peptidergic cells involved in regulation of the CA and reproductive organs in the silkworm Bombyx mori. We will analyze neurons directly innervating these organs, as well as endocrine Inka cells that may control these organs via humoral pathways. We will employ techniques of molecular biology and immunohistochemistry to identify signaling molecules and peptidergic cells, while transgenic approaches in combination with physiological and behavioral assays will be used to determine roles of specific neuropeptides and their receptors. Expected results are important for understanding of molecular mechanisms underlying regulation of development and reproduction, which require complex neuroendocrine communication between peripheral organs and CNS.

Annotation: Ticks are very important vectors of dangerous pathogens, but regulatory mechanisms required for their successful transmission into the host are poorly understood. Likewise, physiological processes controlling function of the salivary glands, gut and gonads during their life cycle have not been elucidated. Our published and preliminary data indicate that several specific peptidergic neurons innervate these organs which are the most important reservoirs of pathogens. Our unpublished experiments suggest that identified neurons control activity of these organs and associated transmission of pathogens during tick feeding on the host. In this project we will use a combination of techniques (immunohistochemistry, molecular biology and bioinformatics) to identify regulatory molecules (neuropeptide and their receptors) and neuronal pathways important for normal function ofthe salivary glands, digestive system and reproductive organs of the ticks Ixodes ricinus and I. scapularis. We also propose to study the role of specific neuropeptides and their receptors in transmission of pathogens(Rickettsia helvetica and Borrelia burgdorferi) from these tick organs into laboratory mice using RNAi techniques. Expected results may provide tools for effective suppression of pathogen transmission and development of procedures to inhibit reproductive capability of ticks as dangerous ectoparasites of humans and animals.

Annotation: Neuropeptides play a central role in the control of development, reproduction, behavior, feeding and many other physiological processes in Metazoa. In this project we use bioinformatics to identify new genes encoding neuropeptides and in situ hybridization (ISH) to confirm their expression in the silkworm (Bombyx mori). Next we will analyze spatial expression of all known neuropeptide genes using ISH, fluorescent immunohistochemistry and targeted expression of green fluorescent protein. We aim at detection of neuropeptides in the central and peripheral neurons and in the endocrine midgut cells in larval, pupal and adult stages. This extensive analysis yield first data on expression patterns of genes encoding novel neuropeptides and first descriptions of 15 peptidergic systems signaling insect neuropeptides in Lepidoptera. Our results will be published in a monograph describing peptidergic cells in B. mori. This comprehensive work will provide basis for functional studies of more than 40 neuropeptide genes.

Annotation: Ticks are important vectors of pathogens that cause animal and human diseases. The most of tick-bornediseases are characterized by the phenomenon of natural focality. In natural foci, the infection can spread inclose proximity to humans, as evidenced by an increase in urban and suburban foci. The relationships betweenthe pathogenic agents of diseases and their vectors co-evolved under appropriate environmental conditions. Therelationship between tick-borne pathogens are studied at the level pathogen – host, pathogen – vector and vector– host. The interactions between pathogen, tick and host represent a triangle with three interfaces: pathogeninfection of host, pathogen infection of tick and tick feeding on a host. Our interest focuses on the relationshipsbetween vectors and their hosts, as well as among microorganisms in mixed infections, which play an importantrole in the of the pathogen circulation in natural foci.

Annotation: We will study reactions of innate and acquired immunity of European bee-eater (Merops apiaster) chicks on experimentally induced changes in abundance of the haematophagous ectoparasite (Carnus hemapterus) and sapro- or/and necrophagous dipteran larvae. They are abundant in nest cavities of bee-eater situated on end of more than 1 meter long galleries in loamy-sandy perpendicular walls. Their influence on development of bee-eater chicks is not sufficiently investigated. In some nests we will experimentally increase or reduce abundance of C. hemapterus, while in others abundance of sapro- or/and necrophagous larvae. In a part of the chicks we will apply phytohaemagglutinin in the wing webbing and sheep red blood cells into peritoneum. We will study the immunity response – skin-swelling, forming of immunoglobulines, chick condition, sedimentation and haematocrit of differently aged chicks in order also to find out differences in their reaction in dependence on their age.

Annotation: The project focuses on a multidisciplinary analysis of macroinvertebrates from alpine ponds in the Tatra Mts. To achieve objectives of the project, study of 60 ponds is expected. These are an important and irreplaceable part of (sub)alpine ecosystems and may, in terms of effective nature protection, serve as an early warning systems and biotopes suitable for long-term monitoring. In this project various methods and approaches of data processing will be used, combination of which will provide a unique knowledge in respective field.Expected outcomes will provide a precise picture of macroinvertebrates structure of these vulnerable biotopes sensitive to environmental changes, allow detection of impact of various env. gradients, and prediction of changes in zoocoenoses depending on changes in the environment. Genetic structure of model species populations allow to identify centers of diversity, main factors affecting variability and indicates trends in dispersal of species in case of expected env. changes.

Annotation: The project is focused on comparison of species diversity of different types of isolated aquatic habitats in lowland areas of Slovakia. The artificially dig pits in the landscape are unique ecosystems offering a valuable possibility to study colonization of a new habitat in time, succession toward the climax and the subsequent decline of species. The principles of island ecology can be used for evaluation of species diversity of planktonic Crustaceans and molluscs in isolated water bodies. The investigation will run in several isolated aquatic habitats of different age, size and isolation degree in the landscape. As reference localities, we will use relatively old material pits situated out of the floodplains, which had arisen in the past at exploitation of the material for dike construction.

Annotation: The proposed project aims to study taxonomy and phylogeny of genera Roraima Kodada & Jäch and Neblinagena Spangler (Coleoptera, Elmidae). Yet the genus Roraima is known only from the Venezuelan table mountains (tepuis), species of Neblinagena, also known only from Venezuela, inhabit streams in lower altitudes. The main goals of the project are: (1) clarifying species classification together with its formal description, determination of morphological and genetic variability, including possible redefinition of the taxonomic concept of the genus Roraima, (2) identification of larval stages using molecular markers, assigning larvae to the species and their description using morphological characters, (3) analysis and interpretation of phylogeny of the genus Roraima, drawing possible scenarios of the speciation and geographic distribution of species in the environment of the table mountains and testing hypotheses of the evolution of the table mountains biota using molecular data, (4) analysis of the phylogenetic position of Roraima and Neblinagena within subfamily Larainae based on combination of both morphological and genetic characters.

Utilization of transgenic approaches for functional analysis of neuropeptides and their receptors regulating behavior and development in insects

Annotation: In this project we will utilize methods of molecular biology, transgenic approaches and physiological experiments to elucidate mechanisms of action of neuropeptides and their receptors necessary for development and behavior in the model species (Bombyx mori). We will focus on identification of complex neuroendocrine interactions between ecdysis triggering hormones (ETH) and their target neurons in the CNS and endocrine organs producing ETH receptors (ETHR). We will use techniques of immunohistochemistry and in situ hybridization for identification of the target cells expressing ETHR. Molecular methods will be used for identification of genes encoding neuropeptides that are expressed in the ETHR neurons and endocrine cells. We will utilize the baculovirus expression system for identification of promoters for specific neuropeptide genes that will be used for targeted expression of molecular markers in the ETHR cells. Transgenic lines producing those molecular markers in the target ETHR neurons and organs will be used for functional analysis in behavioral assays in vivo and for Ca2+ imaging and electrophysiology in vitro. Anticipated results are important for understanding of molecular mechanisms for behavior and development that require complex neuroendocrine regulation between peripheral endocrine organs and the CNS.

The main output of the project will be biological, respectively ecological and biological evaluation and conservation categorization of channels in the study area, which will serve as a basis for their protection and conservation biology management.

Annotation: Intraspecific variability of Anaplasma phagocytophilum in Slovakia is investigated. DNA sequences of differentstrains are analyzed using the molecular methods to assess the genetic variability and the presence of differentgenotypes in ticks and hosts. The occurence and circulation of different genotypes in the natural foci andlivestock is studied. Their associations to the specific reservoir hosts and vectors in the natural foci will beelucidated, that will have implication in the ecology of this pathogen also in other European countries. Project isoriented to a basic research with potential output in the veterinary practice of livestock.

Annotation: The project includes field, experimental and molecular-biological approach and acoustic analyses of study of differences of different behavioral and reproduction parameters of reed warbler (Acrocephalus scirpaeus) as a the model species that nests in two different microhabitats in cat tail (Typha sp.) and reed (Phragmites australis) in the same locality. Exerimentally and in natural condition we will study possible difference in degree of predation of reed warbler nests in two types of stds and its possible correlation with density of nests of other passeriform species of the genera Acrocephalus, Emberiza and Panurus. In the cat-tail and reed stand we will search the potential differences in alarm calls on different species of predators in different distance from the nests We will study, whether difference do exist in morphological and condition parameters of adults and chicks of reed warbler and by means of DNA analyses differences in frequency of estra-pair chicks in nests in cat tail and reed stands.

Annotation: The project aim is to determine interactions between land use practices and water and riparian biota in mountainand submountain streams of Slovakia. Natural habitats of mountain streams flowing through the Tatra NationalPark together with habitats affected by the wind calamity have been selected as a model research objects.Another model objects will be submountain rivers flowing through different types of forests and partiallydeforested landscape. A wide range of aquatic organisms (phytobenthos, meiobenthos, micro- andmacrozoobenthos) and selected groups of riparian flora (macrophytes, vascular plants) and fauna (beetles, birds)and their responses to different types and intensity of land use practices will be studied. Together with assessingof ecological metrics and their responses to studied factors, this approach will allow a complex view on changesin aquatic ecosystems. The project will assess the trends in the formation of aquatic and semiaquaticcommunities in space and time of river networks.

What are the driving forces for the use of inshore riverine habitats by fish species?

Annotation: The proposed project is dealing with the phenomenon of diel dynamics of fish species between offshore and inshore habitats. On the model area of six lowland rivers in Slovakia, we will test three hypotheses whether the use of the inshore area is driven by environmental gradients, feeding activity or predator avoidance. We will follow the variability of environmental variables during the diel period that could be important for fish energetic costs. Also feeding activity connected with the activity of prey items or occurrence of predators could have an important role in diel distribution patterns. We will follow the diel dynamics of net-zooplankton and drifting organisms and the diversity of macroinvertebrates for the purposes of stomach contents analyses. Additionally, we will analyse the intensity of infection of random representative samples to avoid the fish behaviour changes caused by parasites. The side effect of the project includes recommendations for the lowland river monitoring programmes of fishes.

Annotation: According to recent clinical studies, honey as a natural antimicrobial product represents a potential wound healing agent in patients with chronic wounds. The most of serious human non-healing infections accompanied with higher antimicrobial resistance can be characterised by bioflm formation. The aim of present study is to investigate the honey-induced elimination process of biofilm in selelcted multi-resistant clinical strains islated from chronic wounds. Several slovak honey together with medical grade honey and will be used in the study. The honey compound(s) responsible for disintegration and disruption of bacterial biofilm will be identified and isolated from the most effective honey. During the study, the effect of honey on induction of expression of antimicrobial peptides from human keratinocytes will be investigated.

Annotation: The aim of the project is to develop oligo-chip that will detect all relevant tick-borne bacteria present in central Europe in one step. To design the oligonucleotide probes, it is necessary to know DNA sequences of the microorganisms that will be targeted by DNA-chip. We will type all new Borrelia burgdorferi s.l. strains as well as members of the family Anaplasmataceae by the PCR-based methods. Representative samples of each type will be sequenced and oligonucleotide probes will be designed. Probes for the known species will be designed by the comparisions of these sequences available in the Genbank. Implementation of nested PCR will increase the sensitivity of the oligo-chip. Final DNA-chip will be tested on a sample of experimentaly infected ticks as well as ticks collected from the nature. Sensitivity, specificity and discriminatory power of the DNA-chip with the conventional detection methods will be compared and evaluated
