Biotick\u00e9 interakcie zohr\u00e1vaj\u00fa d\u00f4le\u017eit\u00fa \u00falohu pri \u0161trukt\u00farovan\u00ed modern\u00fdch spolo\u010denstiev, ale priame d\u00f4kazy o biotick\u00fdch interakci\u00e1ch s\u00fa vo fos\u00edlnych z\u00e1znamoch do zna\u010dnej miery prehliadan\u00e9 a v\u00e4\u010d\u0161inou sa obmedzuj\u00fa na jant\u00e1r s nieko\u013ek\u00fdmi v\u00fdnimkami. \u0160v\u00e1by s\u00fa ide\u00e1lnou skupinou hmyzu na tak\u00e9to \u0161t\u00fadium, preto\u017ee s\u00fa bohat\u00e9 na paleozoick\u00e9 a druhohorn\u00e9 fos\u00edlne z\u00e1znamy s viac ako 110 000 vzorkami a mnoh\u00e9 maj\u00fa v\u00fdnimo\u010dn\u00fd stav zachovania. Medzi \u0161v\u00e1bmi existuj\u00fa r\u00f4zne typy interakci\u00ed s rastlinami, hubami, eubakt\u00e9riami, archaeabakteriami a r\u00f4znymi \u017eivo\u010d\u00edchmi. V r\u00e1mci projektu sa bud\u00fa \u0161tudovat 1) ektosymbiotick\u00e9j asoci\u00e1cie \u0161v\u00e1bov s fos\u00edlnymi rozto\u010dmi v ~ 130 mili\u00f3nov rokov starom jant\u00e1ri z Libanonu; 2) endosymbiotick\u00e9 asoci\u00e1cie s mo\u017en\u00fdmi bakt\u00e9riami a interakcie medzi rastlinami a \u0161v\u00e1bmi na z\u00e1klade anal\u00fdzy obsahu \u010driev a fos\u00edlneho trusu v ~98 mili\u00f3nov rokov starom kachinskom jant\u00e1ri a ~50 mili\u00f3nov rokov star\u00fdchsedimentoch z Green River, USA; 3) ekol\u00f3gia a mo\u017ene intra\u0161pecifick\u00e9 spr\u00e1vanie vyhynut\u00fdch \u0161v\u00e1bov na z\u00e1klade mikro\u0161trukt\u00far zmyslov\u00fdch org\u00e1nov v r\u00f4znych druhohorn\u00fdch jant\u00e1roch. Tento jav je zn\u00e1my ako metabolick\u00e9 programovanie. Hoci bol tento fenom\u00f3n pozorovan\u00fd u r\u00f4znych druhoch, vr\u00e1tane drozofily, jeho molekul\u00e1rne mechanizmy nie s\u00fa zn\u00e1me. Predkladan\u00fd projekt sa zameriava na \u00falohu mikroRNA (mal\u00e9, nek\u00f3duj\u00face RNA molekuly, ktor\u00e9 p\u00f4sobia ako post-transkrip\u010dn\u00e9 regul\u00e1tory) v metabolickom programovan\u00ed u modelov\u00e9ho organizmu Drosophila melanogaster. Projekt m\u00e1 tri hlavn\u00e9 ciele:1) Identifikova\u0165 hlavn\u00e9 nutri\u010dn\u00e9 komponenty potravy prij\u00edmanej po\u010das v\u00fdvinu, ktor\u00e9 s\u00fa zapojen\u00e9 do metabolick\u00e9ho programovania; 2) Ur\u010di\u0165 k\u013e\u00fa\u010dov\u00e9 mikroRNA, ktor\u00e9 s\u00fa ovplyvnen\u00e9 potravou a s\u00fa nevyhnutn\u00e9 pre dlhodob\u00e9zmeny v mno\u017estve energetick\u00fdch metabolitov (tuk, glykog\u00e9n, gluk\u00f3za, trehal\u00f3za); 3) Odhali\u0165 konkr\u00e9tne cie\u013eov\u00e9 g\u00e9ny t\u00fdchto mikroRNA, ktor\u00e9 prispievaj\u00fa k metabolick\u00fdm zmen\u00e1m. Inovat\u00edvne bud\u00fa pou\u017eit\u00e9 dlhodob\u00e9 d\u00e1ta o pomeroch pohlav\u00ed ml\u00e1\u010fat vt\u00e1kov ako potencion\u00e1lnom indik\u00e1tore kvality prostredia, a v\u00fdstavba energetickej infra\u0161trukt\u00fary sa vyu\u017eije pre lep\u0161ie pochopenie distrib\u00facie modelov\u00fdch \u017eivo\u010d\u00edchov pr\u00edstupom pr\u00edrodn\u00e9ho resp. kv\u00e1zi-experimentu. Preva\u017ene dlhodob\u00e9 d\u00e1ta sa bud\u00fa analyzova\u0165 pomocou robustn\u00fdch \u0161tatistick\u00fdch modelov. Ich \u00fapln\u00e1 ekologick\u00e1 a evolu\u010dn\u00e1 hist\u00f3ria v\u0161ak zost\u00e1va nejasn\u00e1. Tento projekt sk\u00fama fos\u00edlne \u0161v\u00e1byprostredn\u00edctvom: 1) systematiky, 2) taxon\u00f3mie, 3) tafon\u00f3mie, 4) paleoekol\u00f3gie a 5) paleogeografie \u2013 sozameran\u00edm na diverzifik\u00e1ciu, vymieranie, biotick\u00e9 interakcie a morfologick\u00fa variabilitu. Sk\u00faman\u00fd materi\u00e1l budepoch\u00e1dza\u0165 z paleozoick\u00fdch sedimentov (~323\u2013298 Ma), mezozoick\u00e9ho jant\u00e1ru a sedimentov (~251\u201366 Ma), akoaj z modern\u00fdch da\u017e\u010fov\u00fdch pralesov. Viacer\u00e9 aspekty z\u00e1kladnej biol\u00f3gie a fenol\u00f3gie druhu s\u00fa st\u00e1le nepresk\u00faman\u00e9. Glob\u00e1lne zmeny v \u017eivotnom prostred\u00ed vr\u00e1tane klimatick\u00fdch zmien, zmeny charakteru krajiny a urbaniz\u00e1cie sa podie\u013eaj\u00fa nielen na zmen\u00e1ch v distrib\u00facii klie\u0161\u0165ov, ale ovplyv\u0148uj\u00fa aj bion\u00f3miu a sez\u00f3nnu aktivitu klie\u0161\u0165ov. Pijak lu\u017en\u00fd, Dermacentor reticulatus sa meniacim sa podmienkam r\u00fdchlo prisp\u00f4sobuje a jeho are\u00e1l roz\u0161\u00edrenia sa zv\u00e4\u010d\u0161uje. Je potrebn\u00e9 podrobne presk\u00fama\u0165 oblasti, kde sa tieto klie\u0161te vyskytuj\u00fa (pr\u00edrodn\u00e9 aj mestsk\u00e9 biotopy), ke\u010f\u017ee ich are\u00e1l v\u00fdskytu sa v priebehu posledn\u00fdch desa\u0165ro\u010d\u00ed zm enil. Hustota klie\u0161\u0165ov infikovan\u00fdch patog\u00e9nnymi agens je hlavn\u00fdm rizikov\u00fdm faktorom pre \u013eud\u00ed, ktor\u00ed s\u00fa v danej oblasti vystaven\u00ed klie\u0161\u0165om. V navrhovanom projekte bude sledovan\u00e1 aktivita klie\u0161\u0165ov \u010d\u00edhaj\u00facich na hostite\u013ea pomocou met\u00f3dy \u201eklie\u0161\u0165ov\u00fdch z\u00e1hradiek\u201c, ale aj prostredn\u00edctvom zberu klie\u0161\u0165ov z veget\u00e1cie met\u00f3dou vlajkovania. Pomocou metodiky \u201eklie\u0161\u0165ov\u00fdch z\u00e1hradiek\u201c budeme sledova\u0165 aj \u017eivotn\u00fd cyklus klie\u0161\u0165ov a sez\u00f3nnu aktivitu jednotliv\u00fdch v\u00fdvojov\u00fdch \u0161t\u00e1di\u00ed, ako aj ich pre\u017e\u00edvanie. Ke\u010f\u017ee tieto dva druhy klie\u0161\u0165ov s\u00fa pova\u017eovan\u00e9 za epidemiologicky najv\u00fdznamnej\u0161ie, zameriame sa aj na prevalenciu a v\u00fdskyt klie\u0161\u0165ov infikovan\u00fdch patog\u00e9nnymi agens nielen v pr\u00edpade nenacican\u00fdch klie\u0161\u0165ov, ale aj klie\u0161\u0165ov cicaj\u00facich na zvierat\u00e1ch. V d\u00f4sledku meniacich sa podmienok, budeme sledova\u0165 aj prenikanie a v\u00fdskyt nep\u00f4vodn\u00fdch druhov klie\u0161\u0165ov na Slovensku, ke\u010f\u017ee tieto druhy klie\u0161\u0165ov m\u00f4\u017eu do na\u0161ej oblasti zan\u00e1\u0161a\u0165 nov\u00e9 patog\u00e9ny.
